For enrichment of Vibrio species. ALKALINE PEPTONE WATER (VEG.) is a pre-enrichment medium specially standardized for Vibrio species. Clinical samples like swabs and faeces in food and water samples can be added directly to the medium. Veg. Peptone provides nitrogenous, carbonaceous, and other important nutrients. Addition of 10 grams of seafood to 90ml of Alkaline Peptone Water and incubate it for up to 18-20 hours at 37°C. If the incubation period would get prolonged, it causes the suppressed contaminating organisms to develop. The high concentration of sodium chloride promotes the growth of Vibrio cholerae whereas the relatively high pH value of the medium suppresses the accompanying microbial flora at 18-24 hours at 35°C.

Vibrio is a genus of Gram negative bacteria facultative anaerobes that test positive for oxidase and do not form spores. Vibrio species is mainly responsible for causing cholera and food poisoning in humans. Vibrio cholera causes cholerae due to the intake of contaminated food. Vibrio parahaemolyticus is a major cause of food borne infections, causing food poisoning. Since Vibrio species naturally occur in sea water, there is a need for sodium chloride, although some species can grow with minimum sodium chloride concentration. The widely used media for Vibrio isolation are TCBS Agar and Alkaline Peptone Water. Alkaline Saline Peptone Water (ASPW) was first formulated by Shread, Donovan and Lee to be used as a non-selective enrichment broth for the cultivation of Aeromonas species. Alkaline Saline Peptone Water (ASPW) is in accordance with ISO/TS 21872-1:2007 which specifies a horizontal method for the detection of the two main pathogenic Vibrio species causing intestinal illness in humans: V. parahaemolyticus and V. cholera. It is applicable to products intended for human consumption and the feeding of animals, and environmental samples in the area of food production and food handling. Peptone provides amino acids and nitrogen for growth requirements, whereas sodium chloride provides essential electrolytes for maintenance of the osmotic balance. Peptone Water uses elevated pH and salt levels to provide a favorable environment for enrichment of Vibrio spa and also has an inhibitory action on the accompanying microflora.

However, it is intended for the examination of clear liquid or water-soluble materials. Fluid Thioglycollate Medium is also routinely used to check the sterility of stored blood in blood banks. The medium contains Vegetable hydrolysate and Yeast extract those provide the nitrogen and essential growth enhancers necessary for bacterial multiplication. Sodium thioglycollate and L- Cystine acts as a reducing agent which maintains a low oxygen tension by removing molecular oxygen from the environment. Dextrose is a fermentable energy source. On incubation period of 30 - 350 C for 48- 72 hours shown luxuriant growth in all the culture strains tested. Sodium chloride helps maintaining the osmotic balance. The NIH Thioglycollate Medium without Indicator is suitable for diagnostic work of viscous products because the lack of indicator avoids possible toxicity to organisms.

For microbiological assay of antibiotics. ANTIBIOTIC ASSAY MEDIUM NO. 3 (ASSAY BROTH) (VEG.) is used for microbiological assay of antibiotics. This medium recommended by USP and FDA for the Turbidimetric antibiotic assay of Amikacin, Bacitracin, Kanamycin, Streptomycin and other antibiotics. Turbidimetric antibiotic assay is based on the change or inhibition of growth of a test microorganism in a liquid medium containing a uniform concentration of an antibiotic. After incubation of the test organism in the working dilutions of the antibiotics, the amount of growth is determined by measuring the light transmittance using spectrophotometer. The concentration of antibiotic is determined by comparing amounts of growth obtained with that given by the reference standard solutions. Use of this method is appropriate only when test samples are clear. Veg. assay broth composed of Veg. peptone, Veg. extract (nitrogen, amino acid), Yeast extract (source of Vitamin B complex), Sodium chloride (osmotic equilibrium), Phosphates (buffer) and Dextrose (carbon and energy source).
