Product Description
Trypsin is a serine hydrolase that can cut off the carboxyl terminus of lysine and arginine in polypeptide chains. Trypsin digests are used in tissue cell extraction, culture, and in vitro cell culture to hydrolyse intercellular proteins and dissociate tissues or cells into individual cells. Conventional trypsin is obtained from the pancreas of animals such as pigs and cows. The origin of conventional pancreatic enzymes and the characteristics of the production process mean that they inevitably contain impurities of animal origin and potential viral contamination. This product uses recombinant porcine trypsin to replace conventional trypsin. No animal-derived raw materials are used in the production process, and the purity and viability of the recombinant porcine trypsin is ensured through multiple rounds of purification. This allows it to not only replace the function of conventional trypsin, but also to be used for the digestion of special cells or tissues where animal-free origin is strictly required. In addition the product contains EDTA and phenol red indicators.
Storage and Shipping Conditions
Ship with wet ice; Store at 4 away from light for 12 months. If the product is not used for a short time, it is recommended to store at -20 for a longer time.

Assay Protocol
Note : The following protocols are mainly used for adherent cells. For tissue digestion, the tissue needs to be cut into appropriate sizes first. The time required for digestion varies greatly from tissue to tissue, and it is usually advisable for tissues to be sufficiently broken up after digestion.
1. Remove the original cell culture medium and wash the cells 1-2 times with PBS, serum-free medium or other buffers to remove residual serum;
2. Add the appropriate amount of recombinant trypsin digestion solution to cover the cells and digest under the microscope to observe, most cells digestion time between 30s-3min; for indigestible cells, can be placed in the cell culture incubator to digest for a certain period of time (digestion time is for reference only, the specific digestion time should be determined according to the actual situation).
3. When cells show significant shrinkage or morphological changes, aspirate the reconstituted trypsin digest and add an appropriate amount of serum-containing cell culture medium to terminate the digestion (if the cells are difficult to dislodge from the bottom of the plate when gently blown with a pipetteor, this indicates that the digestion time is insufficient and the digestion can be resumed by adding trypsin cell digest. Avoid over-digestion).
4. Collect cell suspension to a centrifuge tube, centrifuge at 800-1200 rpm/min for 3-5 min, add appropriate amount of serum-containing cell culture medium(optional).
5. Blow cells gently to make cell suspension, which can be used for passaging or subsequent experiments.
Note
1. This product can be directly used for cell digestion at 4°C. The digestion temperature can be selected at room temperature or in an incubator as needed, but please do not preheat the product directly at 37°C. The repeated rise and fall of temperature has a certain effect on enzyme activity.
2. After receipt the product can be stored in portions as required; if need to store at -20°C, avoid repeated freezing and thawing.
3. The digestion time varies between cell lines or tissues, so please check the optimum digestion time to avoid excessive digestion for too long, which may affect cell activity and condition.
4. This product is aseptic and read-to-use, and has been tested for mycoplasma and endotoxin. Please pay attention to aseptic operation during use.
5. For your health and safety, please wear the lab coat and gloves when operating.
