Alizarin Red S alias: Sodium alizarin sulfonate/Alizarin carmine/Sodium 1,2-dihydroxyanthraquinone 3-sulfonate/Alizarin Red S/Alizarin S/Medium Red 3/Alizarin red S sodium salt.

Alizarin red: sodium alizarin sulfonate, orange or yellow brown powder.Easily soluble in water, slightly soluble in ethanol, insoluble in benzene and chloroform.1% aqueous solution, pH 2.15, irritant. It can form colored compounds with many metal ions, and can react with zirconium, thorium, aluminum, titanium, beryllium and calcium.

Alizarin red is an anthraquinone dye widely used to detect calcium deposition in cell culture. The aqueous solution is light yellowish brown, yellow after adding acid, and blue purple after adding alkali. It forms red violet or red orange complexes with aluminum, cobalt, beryllium, zirconium, thorium and other metal ions. A certain amount of fluoride can discolor zirconium, thorium and other complexes.

The principle of alizarin red staining is that alizarin red reacts with calcium to produce a dark red colored compound, so that the calcium nodules deposited outside the cells induced by osteogenesis are also dyed dark red.

Assay Protocol / Procedures

The usage and dosage of biochemical reagents are mainly determined according to the customer's experimental purpose and the experimental methods in existing literature or books. The following uses are for reference only.

1. Alizarin red dye solution is used for calcium salt staining in tissues or cells. Preparation method of 2% Alizarin red dye solution: dissolve 10g of Alizarin red in 500mL pure water, stir for dissolution, adjust the pH to 4.2 with 10% ammonium hydroxide, add Alizarin red S dye solution to the section during dyeing, completely cover the tissues, and dye for 5-10 minutes.The dyeing time shall be determined according to the calcium salt content, and it shall be observed under the microscope in time when the calcium salt is dark orange red.

2. Alizarin red can also be prepared into 0.05-0.2% aqueous solution as complexometric titration indicator and acid-base indicator. The first color change range is pH 3.7 (yellow)~5.2 (purple), and the second color change range is pH 10.1 (purple)~12.0 (light yellow).

Note

1. When the reagent solution is on the sample surface, avoid the existence of bubbles, and ensure that the sample surface is covered.

2. The dyeing time of alizarin red is determined according to the content of calcium salt. It can be observed under the microscope. If the calcium salt becomes dark orange, take it out for washing. If the dye solution takes too long, it may be dispersed.
