Product Description/Introduction
Trypan blue is an organic compound.The chemical formula is C34H24N6Na4O14S4. It is often used as a cell reactive dye to detect the integrity of cell membrane and whether cells are alive.Living cells will not be dyed blue, while dead cells will be dyed light blue.Trypan blue can be swallowed by macrophages, so it can be used as a living stain of macrophages.
Normal living cells with intact membrane structure can repel trypan blue and prevent it from entering the cell.However, for cells with lost activity or incomplete cell membrane, the permeability of the cell membrane increases, which can be dyed blue by trypan blue. It is generally believed that the cell membrane integrity is lost, which means that the cell has died, which is contrary to the effect of neutral red.Therefore, trypan blue staining can be used to distinguish living cells from dead cells easily and quickly.Trypan blue is one of the most commonly used staining methods for dead cells identification in tissue and cell culture. Note that the apoptotic bodies are also trypan blue resistant.
On October 27, 2017, the list of carcinogens released by the International Agency for Research on Cancer of the World Health Organization (WHO) was preliminarily collated for reference and Trypan Blue was included in the list of Category 2B carcinogens.

Assay Protocol / Procedures
The usage and dosage of biochemical reagents are mainly determined according to the customer's experimental purpose and the experimental methods in existing literature or books. The following uses are for reference only.
1. In the living cell staining experiment, first prepare 4% trypan blue mother liquor: weigh 4g trypan blue, add a small amount of distilled water to grind, add 0.9% NaCl to 100mL, filter with filter paper, and store at 4.Or directly use 0.9% normal saline or PBS to prepare mother liquor. When using, use normal saline or PBS to dilute the concentration to 0.4% for living cell staining and counting.When staining cells, it is recommended to mix the cell suspension with 0.4% trypan blue solution at 9:1, mix and dye for 3-5 min, and count the living cells and dead cells under microscope.
2. In trypan blue dyeing of plant leaves, the preparation method of trypan blue dye solution can be referred to:0.02g of trypan blue, 10g of phenol, 10mL of glycerin, 10mL of lactic acid, 10mL of ddH2O are mixed and dissolved. When using, take the above stock solution and ethanol 1:2 to mix and dilute to obtain trypan blue working solution.The leaves to be dyed shall be completely immersed in trypan blue working solution for dyeing, and the dyeing time shall be adjusted according to the plant tenderness. After dyeing, the leaves shall be decolorized and photographed for observation.
Note
1. The dyeing time should not be too long, otherwise the living cells will gradually accumulate dye and dye into color, which will result in low monitoring results;
2. In addition, combined with cell counting method, cell counting and vitality detection can be carried out at the same time.
