Oxolinic acid is a synthetic quinolone antibiotic targeting gram-negative bacteria, especially those responsible for causing urinary tract infections. It has also been reported to be effective against Burkholderia glumae, a bacteria causing grain rot, sheath rot, seedling rot, and bacterial panicle blight. It has been useful in studying transmissible resistance mechanisms of E. coli and S. enterica. It can be used to isolate Gardnerella vaginalis and as a dopamine reuptake inhibitor in neurological research.

Oxolinic acid functions by inhibiting DNA gyrase or topoisomerase II, enzymes essential for DNA synthesis. Oxolinic acid is insoluble in water but is soluble in dilute alkali solutions.

Antibiotics are often used in clinical in vitro tests known as antimicrobial susceptibility tests or ASTs to determine their efficacy against certain bacterial species. They are tested against gram-negative and gram-positive bacteria using panels, discs, and MIC strips by medical microbiologists. ASTs decrease the risk of using an antibiotic against bacteria exhibiting resistance to it, and the results are used in clinical settings to determine which antibiotic(s) to prescribe for various infections.
