Product Description/Introduction
Anti-fade Mounting Medium (with DAPI) is a DAPI-containing water-based tablet sealing reagent used to preserve the fluorescence of tissues and cells. The tablet sealing agent can not only prevent the rapid photobleaching of various common fluorescent dyes, but also complete the DAPI staining of the nucleus at the same time. DAPI is 4', 6-diaminyl-2-phenylindole, is a blue fluorescent dye that can penetrate the cell membrane, it is mainly used to fix the staining of cells, and can produce more than 20 times stronger fluorescence than DAPI itself after combining with double-stranded DNA, without binding to single-stranded DNA. The excitation wavelength of DAPI dye is 340nm, the emission wavelength is 488nm, and the excitation wavelength is 364nm and the emission wavelength is 454nm after combining with double-stranded DNA.
This product contains DAPI, which can be used to complete nuclear staining and sealing at the same time. Just drop a drop of this product on the sample with the pipette gun during sealing and cover the cover glass to complete the operation.

Assay Protocol / Procedures
1. Fixed cell slides, cell smears or tissue sections, etc., are cleaned after staining;
2. After the slices are slightly dried, put a drop of anti-fade Mounting Medium (containing DAPI) on the sample, gently cover the cover glass to avoid bubbles;
3. The tissue sections can then be observed under a fluorescence microscope.
Note
1. Fluorescent substances are prone to quenching. In the case of anti-fade Mounting Medium (containing DAPI), the fluorescence duration can be prolonged and the fluorescence quenching can be slowed down. However, the slide after mounting should be stored at 4 and protected from light. It is recommended to use dark slide box. And take pictures as soon as possible.
2. DAPI is irritating to the human body. For your health and safety, please wear a lab coat and disposable gloves when operating.
