Source: Anabaena flos-aquae

Unit definition
One unit is defined as the amount of enzyme required for complete digestion of 1 μg bacteriophage λ DNA at 37℃ for 1 hr in a 50 μl reaction mixture.

Reaction condition

  • 1X EzBuffer IV, 37℃
  • 1X FastCut Buffer, 37℃

Thermal inactivation :  65℃

Time : 20 min

Methylation effect

Methylation : Cleavage

dam : Cleavage

dcm : Cleavage

CpG : Cleavage

Product component

  • Afl II
  • 10X EzBuffer IV
  • 10X FastCut Buffer
  • 6X DNA loading Buffer
  • Sterile water

Note
Afl II is not sensitive to dam, dcm or CpG methylation. Less than 50% of cleaved fragments are ligated in 20-μl reaction mixtures containing 100-500 units of T4 DNA ligase.

Figure 1. Restriction enzyme activity
1 μg of substrate DNA was digested with increasing amounts of restriction enzyme for 1 hour under appropriate reaction conditions.
