Description
This product is used for returning blue after HE staining or hematoxylin staining. Hematoxylin is ionic and red in acidic condition. It is blue in alkaline conditions. After hematoxylin staining, tissue sections were differentiated with acidic differentiation solution to remove excessive bound hematoxylin, and hematoxylin appeared red or pink at this time. The differentiation was terminated by washing, and then the hematoxylin was treated with weak alkaline solution to make the hematoxylin appear blue. This process is the blue-back effect. In HE staining, the return of blue after hematoxylin staining is very important to make hematoxylin show its due blue color.
This product hematoxylin blue return solution is weak alkaline solution, mainly containing 0.5% ammonia water, used for hematoxylin staining after blue return.
Storage and Handling Conditions
Store and transport at room temperature, valid for 18 months.

Assay Protocol
Take HE staining as an example:
1. Hematoxylin staining: The dewaxed sections are stained with hematoxylin stain solution for 3-5 minutes and washed with tap water.
2. Differentiation: The sections were fully rinsed with hematoxylin differentiation solution for 2-5 s，tap water running water to rinse well.
3. Back to blue: Hematoxylin back to blue solution for 2-5 s, tap water running water to rinse well.
4. Eosin staining: the sections were dehydrated by 85% and 95% gradient ethanol for 5 min each. Then it was stained in eosin (alcohol-soluble) for 5 min, and dehydrated twice with absolute ethanol for 5 min each.
5. Transparent: The slices are then dehydrated with fresh absolute ethanol for 5 min, the xylene is transparent for 5 min, and the fresh xylene is replaced with transparent for 5 min.
6. Mounting: Add neutral gum dropwise for mounting.
Note
1. Hematoxylin blue returning solution is alkaline. If you need to stain with other acid dyes after using this product, rinse the sections under running water for about 5 min and remove all the blue returning solution.
2. For your safety and health, please wear a lab coat and disposable gloves when operating.
