Description
Eosin Y is an acidic dye used in histology for staining cell plasma. The basic principle is that eosin dissociates into negatively charged anions in water, which can bind to the cationic amino groups of proteins in the cytoplasm and give the cytoplasm a red color.
The molecular weight of Eosin Y used in this product is 649.89, and the concentration is 0.05%. It is mostly used with Hematoxylin staining solution (G1004) for staining tissues or cells, and is used to show the cell structure. After staining, the cell plasma shows different shades of pink to peach color. Compared with G1001 Eosin Y Solution (Alcoholic), tissues or cells stained by this product need to be dehydrated by gradient ethanol before xylene transparency and sealing, and the coloring of tissues or cell plasma is lighter than that of G1001.

Pre-experiment preparation
Provide your own xylene, gradient ethanol, anhydrous ethanol, and neutral gum.
Usage
1. Sample pre-processing
1) For paraffin sections: sections were sequentially deparaffinized by xylene for 10 min, replaced by fresh xylene for 10 min, anhydrous ethanol for 5 min, fresh anhydrous ethanol for 5 min, 90% ethanol for 5 min, 75% ethanol for 5 min, and washed in tap water.
2) For frozen sections: frozen sections stored at -20°C should be allowed to stand for 5-10 min to return to room temperature.
2. Eosin Y stain
The above treated sections were directly stained into Eosin Y Solution (Aqueous) for 4-5 min, and then sequentially dehydrated by 70%, 80%, 95%, and 100% ethanol for about 3 min each. The sections were then dehydrated by fresh anhydrous ethanol for 3 min, transparent in xylene for 5 min, replaced by fresh xylene and transparent again for 5 min, and finally sealed with neutral gum.
Note: If used for restaining after staining such as immunohistochemistry, please perform Eosin Y staining after the other staining is completed.
Note
1. Eosin Y solution (alcoholic) can be used repeatedly for several times, each 100 mL of Staining Solution can be used to stain approximately 150 sections. It is recommended to replace the new dye when the tissue coloring is obviously shallow。
2. Please wear lab coat and disposable gloves during operation.
