Principle
The FSTest Fumonisin Rapid Test Kit is based onimmunochromato-graphic assay. The test cassette has a testing window for the observation of assay running and result reading. The testing window has an invisible T (test) line and a C (control) line before running the assay. When the treated sample was applied intothe sample hole on the device, the liquid will laterally flow throughthe surface of the test strip and react with the pre-coated recombinant antigens. If there is amount of fumonisin in the sample, the intensity of T line will be inhibited. Meanwhile, the Cline should always appear after a sample is applied, which indicates a valid result. By comparison the intensity between two lines, the device can indicate the presence of fumonisin in the sample.

Sample preparation

1. Grain (Rice, Wheat, Oats, Red Rice, Soybeans etc.)
   • Take an appropriate amount of grain and grind it, weigh 1.0 g of sample and add it into a 10 mL centrifuge tube. 
     
   • Add 3 mL of extracting solution, re-cap the tube tightly, vortex for 3 mins to mix, then centrifuge at 4000 rpm for 3 mins. 
     
   • Pipette 100 μL of supernatant into a 2 mL centrifuge tube, add 150 μL of diluent solution, shake to mix. 
2. Corn and corn grinding processing products 
   
   • Take an appropriate amount of sample and grind it, weigh 1 g of sample and add it into a 10 mL centrifuge tube. 
     
   • Add 5 mL of extracting solution, re-cap the tube tightly, vortexfor 3 mins to mix, then centrifuge at 4000 rpm for 3 mins. 
     
   • Pipette 100 μL of supernatant into a 2 mL centrifuge tube, add 900 uL of diluent solution, shake to mix. 
     
   • For corn grinding processing products with LOD 2 mg/kg, pipette 50 μL of mixture from 6.2.3. into a 2 mL centrifuge tube andadd 400 μL of diluent solution, vortex to mix, and ready for the test.

For corn with LOD 4 mg/kg, pipette 50 μL of mixture from 6.2.3. into a 2 mL centrifuge tube and add 900 μL of diluent solution, vortex to mix, and ready for the test.

Procedure & Steps 

1. Before use, ensure the cassette is between 20 to 40. 
   
2. Take the test cassette out of the bag and put it on a flat surface. 
   
3. Pipette 100 μL of sample solution into the sample hole of the test cassette.
